螢火蟲熒光素酶在氧氣、ATP和鎂離子同時存在的條件下，催化熒光素氧化，生成氧化熒光素，同時產(chǎn)生黃綠色光，波長540-600nm。

熒光素酶（luciferase）是自然界中能夠產(chǎn)生生物熒光的酶的總稱。熒光素酶可以催化熒光素氧化成氧化熒光素，在熒光素氧化的過程中，會發(fā)出生物熒光。然后可以通過熒光測定儀測定熒光素氧化過程中釋放的生物熒光。熒光素和熒光素酶這一生物發(fā)光體系，可以極其靈敏、高效地檢測基因的表達，是檢測轉(zhuǎn)錄因子與目的基因啟動子區(qū)DNA相互作用的一種檢測方法。

目前，最有代表性的是從甲蟲中分離得到的螢火蟲熒光素酶（Firefly luciferase）和從海腎中分離得到的海腎熒光素酶（Renilla luciferase）。
